Genentech, a member of the Roche Group, today announced positive results from the pivotal Phase III Krascendo 1 clinical trial evaluating divarasib, an investigational targeted inhibitor, in patients with previously treated KRAS G12C-mutated non-small cell lung cancer (NSCLC). The landmark findings were presented at the 2026 European Society for Medical Oncology (ESMO) Congress and simultaneously published in The New England Journal of Medicine.

The Krascendo 1 trial demonstrated clinically meaningful efficacy across primary and secondary survival endpoints, establishing divarasib as a potent next-generation therapeutic option.Median Overall Survival (mOS) reached 17.7 months with divarasib, compared to 12.0 months observed with existing standard KRAS G12C inhibitors. It demonstrated a 44% reduction in the risk of death versus standard treatment arms and achieved a 42% reduction in the risk of disease progression or death. The tolerability & response exhibited a higher confirmed objective response rate alongside fewer reported severe treatment-related adverse events.

Non-small cell lung cancer harboring the KRAS G12C mutation historically carries a challenging prognosis, often complicated by rapid emergence of therapeutic resistance to early-generation inhibitors. The Krascendo 1 outcomes represent a critical advancement in overcoming these oncogenic drivers through more durable targeted inhibition.

The data reinforce the sustained clinical utility of precision oncology, delivering tailored solutions that meaningfully extend patient survival while improving treatment tolerability. Genentech plans to engage global regulatory authorities to advance divarasib toward regulatory review and expand innovative treatment options for patients worldwide.
